Microbial fructosyltransferase: evaluation of pH and temperature effects on transfructosylation and hydrolytic activities

Abstract:

Extracellular fructosyltransferases (FTase, E.C. 2.4.1.9) from Aspergillus oryzae IPT-301 were investigated for the production of fructooligosaccharides (FOS). These enzymes exhibit transfructosylation (At) and hydrolytic (Ah) activities that can be altered by modifying the reaction conditions. This study aimed at verifying the influence of temperature and pH on FTase activity and the ratio between the activities (At/Ah). The enzymatic activities were evaluated in the temperature range from 30 ⁰C to 65 ⁰C and pH range from 3.0 to 6.0. The optimum pH for At ranged from 4.5 to 6.0, while the optimum temperature was 50 ⁰C. For Ah, the optimum temperature and pH were 60 ⁰C to 65 ⁰C and 3.0 to 3.5, respectively.
