Evaluation of temperature and Pheffects on mycelial fructosyltransferase from aspergillus oryzae IPT-301

Abstract:

Thermal and pH stability of mycelial fructosyltransferase (FTase, E.C. 2.4.1.9) were investigated for the enzyme produced by Aspergillus oryzae IPT-301. This enzyme is applied in fructooligosaccharides (FOS) production and exhibits transfructosylation (At) and hydrolytic (Ah) activities that can be altered by modifying the reaction conditions. The enzymatic activities were evaluated in the temperature range from 30 ⁰C to 65 ⁰C and pH range from 3.0 to 6.0. The optimum temperature for transfructosylation activity ranged from 45 to 55 ⁰C, in which a high activity ratio (At/Ah) was also observed. The optimum pH for At ranged from 4.5 to 6.0, with transfructosylation values varying from 611.05 to 780.79 U/g.
